Summary: Leaf-level msts. Aci and light response curves

I arrived at 9:30 AM. It was a sunny day with a relatively strong breeze. The sky was clear and there was a clear view of Mount Diablo. The alfalfa was at about knee height. I had the machine ready to go, so after warming up the first leaf was measured at 10:00 AM. I measured 5 other leaves from 10 AM-2:30 PM. The first two leaves I selected were on the sun and had relatively low stomatal conductance whereas the last three leaves had been in the shade a bit more and had higher stomatal conductance.In all leaves, I performed ACi curves with the leaf chamber fluorometer.

The CO2 concentration was varied in the following order: 400,300,200,150,100,75,50,400,500,600,800,1000,1300 ppm.

I did light response curves on the last 3 leaves using the following light levels:
2000,1500,1000,500,300,200,100,50,20,0 umol m-2 s-1.

Only 2 leaves were big enough to cover all the chamber, the other 3 leaves cover between 80-90 % of the area in the chamber. After the curves were done, the leaves were harvested and I estimated area covered within the leaf cuvette and measured chlorophyll content. I left the site around 3 PM.
